Selank is a synthetic heptapeptide derived from tuftsin (Thr-Lys-Pro-Arg) stabilized by the addition of the tripeptide Pro-Gly-Pro. Studies (PMID 26228446, PMID 21390649) describe modulation of hippocampal BDNF and NGF expression, as well as potentiation of GABAergic transmission without direct affinity for the GABA-A receptor (unlike benzodiazepines). At the hormonal level, the studies observe a moderate reduction in cortisol under acute stress conditions, modulation of serotonin and norepinephrine, and an increase in endogenous enkephalin. Anxiolytic effects without sedation, antidepressant effects, and immunomodulatory effects (modulation of IL-6 and IFN-α) are documented in preclinical models and Russian clinical studies.
