Melanotan II is a non-selective cyclic analog of α-MSH that activates MC1R, MC3R, MC4R, and MC5R. At the hormonal and neurobiological level, studies (PMID 8637402, PMID 15262693) characterized stimulation of melanogenesis via MC1R (pigmentation), as well as activation of hypothalamic MC3R/MC4R pathways involved in the regulation of appetite (documented anorexigenic effect), energy expenditure, and erectile function (via pro-opiomelanocortin neurons modulating nitric oxide at the paraventricular level). MC4R activation alters leptin secretion and satiety signaling. Effects observed in preclinical models include increased eumelanin, reduced food intake, and stimulation of sexual behaviors in rodents.
