Effects documented in the literature

In phase 1 and 2 clinical studies (NEJM 2023, PMID 37366315), Triple R demonstrated simultaneous activation of the three incretin receptors GLP-1R, GIP-R and GCG-R. This triple agonism results in increased glucose-dependent insulin secretion by pancreatic β-cells, suppression of glucagon, slowed gastric emptying, and activation of energy expenditure via hepatic glucagon receptors. At the hormonal level, the studies observe modulation of the entero-insular axis, a decrease in circulating leptin, and a reduction in HbA1c in the cohorts studied. At the body level, the studies report dose-dependent weight loss (up to −24% at 48 weeks at 12 mg/week), a reduction in visceral adipose tissue measured by MRI, and an improvement in liver markers (reduction of non-alcoholic fatty liver disease).
